Yes, even if you don’t have an item equipped you can still lose it if it’s in your backpack inventory and not somewhere like a chest or fridge

Just make sure you empty yourself of all valuables besides like one necessary weapon and bring a lot of food with you when you go for health and energy and if there’s a lot of monsters on a floor eat to regain health when your health is knocked down to halfway

I would focus on delving before combat levels. Combat levels are mostly useful for speedrun/challenge run strategies, for casual play you don't normally need them too high and will achieve them through normal gameplay. Farming the mobs on floors 21/41/81 while farming copper/iron/gold is a solid semi-casual option though
