Amiibos still need to be invited 3 separate times, even if you have an empty plot.

• Can I choose who the Amiibo villager kicks out?
Yes, just make sure you've spoken to that villager at least once and their house isn't currently being relocated.

Yes you can go to mystery island and choose 3 villagers in the same day before you finish the plots. You’ll be able to move them out once you have 6 or more villagers. One of them will ask to leave except your newest one
