Missions give cards, so technically missions do unlock them, but it ain't any specific mission, it's chance.

Depends on what you mean by "start again". Do you mean rebooting the simulation engine? In either case, I think the answer is yes (again, depending on what you mean), but the second time is faster (the more metabits you have, the more things speed up).
