You can get a fifth without keepsakes if the fourth appears on the door in a shop, and one of the shop items is a fifth.
So I guess theoretically you could get up to 8 with keepsakes, although that interaction is quite unlikely to happen in Tartarus.

If you only have 3 gods in your pool by the time you reach Styx shop, you should in fact be able to get all 8 naturally there. I've only been able to get 7 naturally myself that way, but 8 should be possible in theory.

The only way to actually have boons from all of the gods by the end of the run is to get to Styx with only 3 and find the remaining naturally, since that's the only way you can find all 8 and also have the Sigil of the Dead equipped by the end of the run.
