Ehhh... I feel even at Level 50, the Towel's percentage HP increase isn't going to make... much of a real dent, but I guess you can if you want? As long as your AA is good, and few planes get to the backline she should be fine; just need to watch the battleship 3-star node Kongou and cargo 3-star node Haruna, since they're the only two units who can shell your backline
I'd try Radar/Shell first, put them in and see what happens... but in terms of protecting her, she probably is better off with an HPAAR anyway, at least assuming planes get through

So I'd probably reorganise your fleets into 1 CV, 1 BB boss fleet before evaluating the Chkalov vs Enterprise placement

I'm never too happy doing direct comparisons because all of them bring their own utilities to the table, but...
-Guam brings massive AA and boosts your fleet's AA so that is useful in content demanding AA
-Laffey is especially difficult to kill in content where enemy attacks do high number damage due to how her damage reduction skill works, and she also brings massive AA capability and ASW capability (unlike the rest) courtesy of being a DD
-Anchorage is an extremely difficult to kill tank and provides a little defensive support for your vanguard with her smokescreens
In terms of direct comparison with Brest... which one fits best really does depend on the content of the rest of your fleet and what content you're tackling... no real "one size fits all" answer

Brest is usually avoided in opsi cuz her heal can be detrimental iirc. You should probably use her in chapter 14 and 15 since the plane + torpedo combo makes agir a slightly worse tank than she is usually due to her heavy armor
