1. A Mixed zone is not possible in game yet, because you can only set one type of building in plugin coding,so it's either residential or commercial and can't do both. Unless you are asking for a block/new zone that has a mixed kind of building,you can build a residential next to a commercial building if you do it in manual building.
2. I think one of adutchguy plugins did that, specifically the latest north american buildng pack. Which contains or inspired by new York/Manhattan area buildings.
3. I think in game LRT and metro could do similar affect to that as well. Perhaps using some plugin. Even tho it didn't have a customisable route,if it uses a normal train method,you can use the train management plugin by ian' to make a specific route for it.
