The ingredients were in her memory but after being burned at the stake, only her cubes had them. This is what Nicholas was searching for (before David performed and unsolicited lobotomy)
When Jakob sacrifices her cubes to the lake, in the well, the Lake's water dissolves the cubes, from which the ingredient's essence or physical matter can be removed. After dipping the last cube, the recipe for the elixir is complete. Slowly consuming him, the fire kills Jakob, granting enlightenment to the Eilanders. But the elixir allows someone else to live. A sacrifice had been performed (Jakob), allowing a soul to be enlightened (Jakob, as well).
What isn't so obvious is that the well is connected to the lake, and by extension, its water. Thus the elixir slowly, after many years, dissolved completely within the water. This explains TRT's notice: dipping a brain in water (since this is in RL, Lake's water) bleaches said brain. It washes away, dissolves, the memories in it contained.
Harvey was alive during the plagues, we see him in a secret easter egg, and he might have ended up drinking water infused with a big-enough elixir portion. The same applies to Mr. Toad, (frogs are seen during the 2nd plague) and for Mr. Bat too (The Cave is very near, and its possible a bat wandered around too

It's kind of the opposite. He has a choice or at least as much as any elixir duo has. The way I see it, its not yet decided whether or not Dale or Laura will be the winner (unless you view Seasons and UB taking place after their finale, which would imply that Laura loses the draw)

Since Dale is the most likely to be enlightened, he will become Deva and Laura will be corrupted
Somehow, Dale might "stop" the corrupted guests
And maybe later or in the same moment, Mr. Owl may die, change realms or reincarnate
