It's really hard to call any single joker "the single best one" in this game, because a lot of jokers have tradeoffs or only fulfill one of the victory conditions
It's a lot easier to reason about "The best chip joker" or "The best +Mult jokers", but even then the answer may depend on your build and situation
Stuntman is pretty good, but not every build wants a Stuntman. It's run-winning on Plasma Deck when you get it early but not as good in Ante 7 or 8
Perkeo is probably the closest contender because it amplifies any consumables you get and is one of the most versatile jokers in the game... but it also does nothing initially, and its true power is hard to bring out without Ghost Deck or Seance
I've had runs where it did nothing for 2 antes because the only consumables offered in the shop were Neptune or The Lovers

Any meaningful deck manip is good, and for example you invest 3$ in neptune, and get 3$ every round from perkeo alone

i think the quality of the greater lore in a part of the game is pretty directly proportional to the quality of that part gameplay wise though
