First things first, test with as many cables as you own...
If the quad connects normally (when not trying to flash) then please look for and download the "Impulse RC Driver Fixer" - and also download a program called "Zadig" - these are assuming you are using windows.
i just picked my quad up first time in 3 yrs.. so was expecting some issues lol. i had that message (Failed to open serial port) as well.
turns out thats a very common message, different things cause that message. go through this list, hopefully itll be one of them.

For me.. i got up to running the ImpulseRC Driver Fixer, that gave me an error of 'Too many devices' (or similar)
turns out i had to unplug my x-rsr receiver, then ImpulseRC ran fine, fixed whatever the driver issue was.. and no more 'Failed to open serial port'
but yea a lot of people had way different stuff going on. like hroll said "check cable first, make sure its a data cable, connect with something else to confirm it, phone etc..

@bronze root @iron crescent Often had the same problem. Desolder or disconnect the receiver and than flash FC. It appears to be a problem on certain FC and some times when flashing from one FW generation to a newer one. It's truly annoying.
