No, you generally want to use your Skill when Poison Blast is on cooldown and your Toxified Meter isn't full/sparkling.

You'll see a symbol at the right of the boss's health bar that tells you Poison Blast is on cooldown.

Without any [Moderation] attributes, you want to be particularly picky when you use it. It gives you access to the fastest way to activate a Poison Blast, with the enhanced Aerial Attacks.

During your Ultimate though, you can use it whenever your buff expires and spam Aerial Attacks in between.

Start → Use Skill → Spam Aerial Attacks → Poison Blast happens → Poison Surge activates → Continue spamming Aerial Attacks → Poison Surge deactivates → Repeat from Start

Skill does a quick burst of accumulation alongside buffing your Elemental Accumulation rate slightly. The enhanced Aerial Attacks after will do even more accumulation, allowing you to proc Poison Blast often before the buff expires.

I think there is no real answer for that. The devs simply didn't implement Solo Mode on that...yet.
BUT...you can actually enter it solo. Simply deactivate "Fill Party" and it should start immediately with only you. But the difficulty stays the same as trios. Though it's not that hard if you're a good Showdown player. I mostly do Doom alone too, with Ziping Yin.
