Runes are pretty simple.
If it says it adds debuff to trigger. Then use it on hero with triggers. Any should work. Obviously check if the hero has the said debuff already.
And there is a rune that says that increase it effect. Then you check if your hero have that debuff.

Pretty self explanatory. No need phd degree to figure it out.
