I have an Acer Aspire 3 (Aspire A315-24P). I want to upgrade my BIOS, but it seems that in the drivers webpage there's only an EXE to do it. I was wondering if I can do it using Wine, VirtualBox or if I have to install Windows on my computer. Long answer: boot into a windows ISO and do it

some bioses let you flash an update from within the bios

Dont even think about using that EXE in wine.
Use the non exe BIOS file to do it.
But truthfully, what no one here has mentioned yet is.
DO NOT UPDATE THE BIOS UNLESS YOU HAVE AN ISSUE AND THE PATCH NOTES FOR THE BIOS UPDATE FIX THE ISSUE YOU ARE HAVING

While this is not your issue.
If you build a PC and the motherboard does not support your CPU, a BIOS update is needed to fix it but this means you need a compatible CPU to start with.
This is a valid reason to update the BIOS
Updating for the sake of updating it is not a valid reason.
If you NEED to update the bios, you will need to install windows
You cant just run in the preinstall environment as you will also need to install the chipset drivers from acer to make sure that the BIOS updater can communicate correctly over the SMBUS which may use custom IC chips which will need drivers.
