Pretty sure you have about 15 minutes game time to reach your destination, which is about two minutes real time. That’s plenty of time to get from anywhere to anywhere even in a Very Large City, assuming you’re running and not walking — and you should be running all the time, since there’s no drawbacks to running other than the occasional falling through the floor bug.
If you’re having trouble navigating from point A to point B, here are some tips:

You don’t have to listen to the entire conversation on the phone. In fact, you can hang up as soon as the other party has picked up and started speaking, and your character will automagically know where to go.

Don’t just blindly rely on the floating arrow to guide you, before you begin your journey, open up your map and study the red path so that you can anticipate where to get to. In some cases, you may be able to manually determine a better path than the default suggestion. And don’t worry — time is paused when you’re studying the map, so you’ve got all the time in the word.

Before picking up the side job, if at all possible eat some food and drink either coffee or Starch Kola so that you have the Enegerized, Hydrated, and Focused boosts. At least one of these boosts movements speed by a noticeable amount.

I’ve mentioned this before, but you should always be running! I believe the default key bind is the Shift key.

As a final precaution, always make a save just before picking up the Side Job. You can reload if for some reason you can’t reach your destination in time. If nothing else, it’ll permit you to quickly reload in case the fall through the floor glitch happens to you.
