• fly to a location on the map labelled [Kullervo's Hold]. If it's not visible on the map, you entered on the wrong spiral - it's only there for some of them.
• on the ledge overlooking the arena at the destination, there should be a man sitting on a chair. If you can't talk to anyone there, keep doing the main quest - lack of dialogue means he's part of it. If he can be talked to, talk to him and do the boss fight that he puts you into.

you unlock hardmode by beating every node on the star chart, and unlocks a second star chart to complete for more mastery XP. can swap between normal and steel path content at any time.
enemies become much more durable, significantly more deadly and much more numerous, and a lot of stuff you might've overlooked becomes important to learn fast on it, but you get massively improved rewards overall.

dante's a great choice! I don't know his nuking capability, but he has decent damage output and can keep you alive very easily.

if you want a nuke that struggles to die, kullervo is one. given his second skill's augment, [Volatile Recompense], you'd be hard-pressed to die so long as you stay in melee range of an enemy(which is very easy given his gap-closing skill, wrathful advance, that encourages melee focus), and his third skill turns single-target damage into AoE(and AoE into super AoE)

for mesa's regulators to be optimized, you want the furax wraith as your melee weapon, as it buffs their fire rate via its amalgam mod, as well as a companion with [Reinforced Bond]'s secondary effect active for more fire rate. Both of those bonuses put together handle that part of her regulators' modding, removing the need for it on her gun.
crit chance archon shards on mesa herself also remove the need for crit chance on her guns - without them, [Secondary Enervate] is heavily advised to bump that up. In its place, secondary outburst(which consumes modded-up initial combo from the furax) and secondary fortifier work well.
As far as the gun's modding goes, the thing's a crit-based weapon. Your chosen element being heat lets you rack up arcane hot shot through whatever procs do occur, which helps concentrated fire(pun intended), and the gun gains a serration-like bonus over that same concentration, removing much need for that as well.
As far as helminth subsumes go, it's mostly for energy management, and over her most useless skill - the first one. her other skills increase your offense and defense by an amount that shouldn't be neglected even without explicit focus on them, so they're not worth replacing, and all of her abilities eat energy like crazy with their upkeep, making some sort of support necessary. For stationary mission types like defense, nokko's brightbonnet is the best, allowing mesa to "jump" while buffing her guns' pre-mod damage value and supplying energy mid-channel, and for mobile ones, nourish does something similar to less potency without tying you down to one spot.
Below is what I rock for full optimization of mesa - the furax is overkill, but reinforced bond really does make a difference.

it stalls enemies outside of ability kill range for a little bit and generally hinders aoe effectiveness by keeping enemies spread out, and enemy flow rate is the main speed determiner of defense.

Fissures is where I draw the line. You can inadvertently cause problems with the pacing. And if you're ability strength is too high, you can literally cause everyone to not get their reactant cause you're killing things before they can be corrupted

But Excalibur specializes in melee stuff with his slash dash and exalted blade and has radial blind, which effectively is a survivability mechanic at its core, then there's radial javelin, which (I think) can be used for priming for melees. Cyte buffs guns more directly than Excalibur buffs melees (much more). But both have an exalted weapon, both have something intended for survivability, both have abilities that can help their respective weapon specialization.
