Personally i would suggest treating the media as their own thing, since ultimately it's all different writers doing different things with the story.

Issac isn't necessary for the pre-alpha (the developers are working in 'phases' and Issac is not part of the current one), but is confirmed to be returning in future versions as he is part of the larger story for HN3,
Iris, on the other hand, doesn't have a place in the larger story for hn3. The larger story, though, is all that's present in-game for now, so she isn't in the game. They plan on adding more things that are outside of the main plot for hn3, though, so Iris might be coming back.

Hello Neighbor 2 officially became what the Hello Guest project would turn into, and along with this change

a new storyline was created

and at some point when you're there, Peterson will drive off somewhere and you're meant to follow him

and where he goes is something related to the Outskirts (where A1 and 1.5 take place)

and that's pretty much all I know

oh. actually wait the Hello Neighbor 2 demo takes place in the outskirts too, just that the Guest isn't there and it was probably during the time when the Beta was changing the storyline
