You never need to spend Ambrosia to unlock a character's quest. You just need to give them 6 Nectar to unlock the quest's pre-requisites, then it's RNG to get the quest dialogue to trigger.

Dusa's Companion is unlockable by purchasing enough upgrades for the Lounge back at the House. Just keep upgrading it and at some point her quest will activate. Zag will offer to help her renovate and then you just need a certain number of upgrades bought (the wiki's number is correct I believe).
Skelly's Companion is unlocked by "killing" him with a Lvl 5 Zagreus Aspect Stygius, but he has to ask you to do it first. I didn't have it fully levelled when he asked, but doing it ahead of time is reasonable.

Meg requires lots of converstions with her talking about her and Zag's old relationship, mostly from the Nectar conversations. She also needs to appear in Zagreus' room twice; once to "collect her things" and a second to talk to Zagreus about reconciling. After the second time you can get her Companion next time she's in the House (I think. Been awhile since I got that one).

Sisyphus' quest chain involves a lot of dialogue. Talk to him until Zag expresses wanting to ease his sentence, then you need to talk about it with Hades, Megaera, and Nyx. Than also has something to say but idk if that's related to the quest. Once you have Administrative Privilege, Nyx can tell you in which folder Sisyphus' sentence is - interact with it in the Office. After the next run you can buy it for 7 Diamonds at the Contractor. Then you need to go find Sisyphus to complete the quest. Give him Ambrosia for the Companion.
The Bedroom Conversation at Max Affinity with Than, Meg, and Dusa is the romance decider. With Meg and Than, you can go on as lovers (even at the same time, there's no repercussions. it's Greece after all) or just as friends. With Dusa, you become Besties forever.
I know entirely too much about this game but hey, there is no escape.

Between 2-3 per character. More for a certain one, a lot more.
Also not everything is fully unlocked yet, there's things that can't be done just yet so who knows.
Also remember that ranking up the companions costs Ambrosia.

There's six companions, each costs 10 ambrosia to fully rank. Most of the characters take between 2-3 Ambrosia to forge a bond, there's one that needs... 9 or so?
I used my Ambrosia occasionally to buy some blood, but I'd probably save at least a chunk of it. If you don't have any companion try to keep at least 10 in the pocket to get one maxed out.

for just getting darkness, yeah. going from the most desired rewards to darkness is a bit disappointing. no one said they wanted chaos spots to be common enough that it makes the high tier rewards they give trivial, we just wanted them to not be so obnoxiously hard to find. but alas this is the trade off

Idk why. One interaction I feel like should happen is Hades scolding Hypnos and Hypnos, somehow, misunderstanding the scolding to mean he might be allowed to sleep more/take a nap break. Idk why that's a thing in my head...
However, considering the progress I'm making in the dialogue, I doubt that's a thing. Hypnos sounds like he's finally getting organized and focused.

I can say that in relation to finally getting the dialogue where Dusa gets fired for overworking herself and establishing a personal relationship with Zagreus. Took... two runs to finally get Nyx to comment on it and briefly SHAKE THE HOUSE.

i had the talk with nyx about firing dusa
went into the bedroom, forgot to check merchant, went back out
and there was dusa having the dialouge of being reinstated
