non skyraider db is not preferable on flex slots because it usually sports the least single target dmg compared to the other good planes from other types, and we only have 1 skyraider so it almost never goes to a flex slot either way
now for haku flex slot the same thing applies, conv tb have higher single target dmg, ud only use tenrai if u need splash
for the rest of the setup it depends against what armor type ure going up against

It's unlikely to make too much of a difference as you don't want to push your subs that hard anyway. 99% of the time you use subs for only 20-25s. Any longer and you risk them dying and a dead sub is a useless sub.

unicorn retrofit is the best healer, helena is good later on, portland is a pretty good main tank and is needed for beginner missions, your starter dd is also a pretty good ship and needed for beginner missions
