It is one time use but it grows back after a while. There are 3 levels to it with celestial being the highest which you get for letting it mature on the tree. You can even get a relic for letting it mature but I don't know how long it takes.

If you get 3 celestial (let it mature a few days) you get a free relic heart too

Another pro tip is that you can hold one of each "maturity" at a time

#

so once you already have the ripened heart, what you want to do is have a single mature, elder and celestial fruit in your inventory at a time
