Not necessarily. Some transmission need to be checked while the car is running, like my car.
And some need to be checked after reaching operating temp and going through the process shifting through the gears like you said, but not running.
well it was running while i checked it
Some cars have special fluid top up/change procedures. Also there are some which you can only check when running or at a certain temp.

@viscid lake seems okay, might need a top-up. I would double check either on a mustang only Reddit or check in/if you have a more expanded owners manual.
Do you know if it is to be checked while running, off hot, or off cold?
