For what it's worth I found this on the internet - not sure its right but I'll follow it for now

1 yellow stamina pot (refund 25% stamina) - 2 purple pots (ae attack) - 1 5x pow pot (for when you start getting resisted), 1 5x chance to find grand discoveries.

Character 1 - pops 2x purple, 1x yellow pot, and plows down to floor 80ish
character 2 - once it is mostly 'final object' pop 5x discovery
character 3- once you start getting resisted pop the green power potions

too broad a question, pretty much every mechanic in the game has a way to increase your damage, go through them one by one and look where you can improve
