Once you unlock each mastery, and assuming you've got funeral flowers unlocked, it's a good idea to backtrack floors and improve your gear and charm before you hop to the newly unlocked mastery. If you hop too soon without refreshing your stuff, you'll inevitably feel the need to backtrack regardless but this time you won't have your funeral flowers multiplier since it resets upon changing mastery

Once you enter new floor you put everyone with yellow belts. Once you feel your funereal flower multi is enough, on 1 char pick rosarie, silk veil and nunchaku on rest pick lotus flowers. This setup will allow you to push easly through masteries.
