Here's how I do a standard winning run:
Cover as much as I can on each stage, picking up every item I can find
But don't retrace your steps too much or you'll just waste time
On stage 4, GET THE RED ITEM, if it's in one of the expensive chests, check the chest to see how much it costs, and then farm a little and maybe do the teleporter if you have close to enough
If you're on sirens call, then hunt down the eggs and break them, I think you need to break 5
Then eventually get to commencement, do the pillars (pillars of blood are the easiest) and head on up there
