Right one is the best as that leaves all the air at the top of the radiator away from the tubes
how ever realisticly speaking it doesn't matter that much usually

Air in blocks shouldn't be a issue if it's designed with a good pump
it should just push it out
but it's not entirely stupid to think off tbh

the best would be if the block is underneath the cooler and also have the pipe connecting as low as possible
