You can format the drive to wipe it clean of all partitions, then create the partitions how you like.

Formatting requires wiping the drive so... both. You might be able to get away with just wiping the latter partition, then using Windows Disk Management to extend the first partition though.

Regardless, you should always have a backup of any data you cannot easily re-create / download again.
