My theory is that it's detecting a truffle worm, which is supposed to be extremely rare.

JzBoy said the next boss will be Duke Fishron, and Navashark is the fisher of the group. It'll make sense for him to really want the truffle worm for the 666% fishing power.

I think Koonboat just really hates anything that isn't orange juice. He does use a magic mirror as seen in the Queen Slime episode, he's mad that Navashark tricked him into drinking a recall potion.
