i'm not actually sure. if we look further back, the first time he was actually exposed to the possibility was when polnareff mentioned the oddity of being reversed when he was trying to climb the stairs towards dio in his mansion. jotoro could've inferred what the stand ability was but just kept quiet, since it's a known trait of his to just stay quiet and always assume everybody knows what he's thinking.
