Okay just looked up all of Slayer's moves and abilities in Guilty Gear Xrd and he doesnt seem to do anything particularly special. The teleport thing is actually something that Part 1 Dio was shown to do in the anime and the rest of what he does is generic Vampire stuff like health drain and hitting hard and fast. To determine if he would beat part 1 Dio I would need to look at each characters feats of srtength and speed to see how they match up, but my gut feeling is that Dio's Vamp-eye-r lasers and freeze touch would probably be enough to stop Slayer in his tracks. as for DIO, Slayer has no means to counter The World.

the most common answer for the DIO thing is the whole "this isn't my body" thing, since he can't heal either, he can't evaporate the moisture of the body nor control the blood flow throw his eyes
also since he can't heal properly, it would be very bad for him to shoot out the lazer thing
as why he doesn't do that after driking the blood, no answer there
he just doesn't do it, could be because he thinks he doesn't need it since he has the world
