Also, the slope is negative, because the line is going down as ñ is going to the right.
In a Cartesian plane, the slope of a line is given by m = (Y_A - Y_B) / (X_A - X_B);
If the function representing the line is increasing when X_A < X_B, then Y_A < Y_B;
If the function is decreasing when X_A < X_B => Y_A > Y_B; (Y changes sign)
This results into a negative sign in m = (Y_A - Y_B) / (X_A - X_B);

Anyway, here is the idea: you want to know why when n=ñ, ñ/[L] gives a different result than n-ñ / K_d = 0;
well, n=ñ happenes ONLY when ñ/[L] = 0, in other words, the equality is available ONLY when ñ=0 (0/[L] = 0-0 /K_d)
It doesn't apply to any ñ value, because the x-intercept value is tied to the relation f(x) = 0; If one works, the other one will too; If one fails, the other one will follow;

if n = 100, then n-ñ = 0, but ñ/[L] equals 100/[L] not 0, which is why i'm confused

As an idea, it all comes down to you extract data from the graph, and the smalles mistake in the algrebra can get to the wrong answer
