"Quand est-ce que les enfants ouvrent les cadeaux " is good in french. You don't need to replace something.
But the sentence : *quand est ce que les enfants LES ouvrent"
Is also good in french. Here, you replace : Les cadeaux
By : les

here des is considered an article, not de + les
En in this case would mean they open "some" but not all of their presents, basically
