Well first of all you need to know what those four pronouns represent before you know when to use them

Y -> replacing à but not human or loca
Je suis à l'école -> j'y suis
Je joue au tennis -> j'y joue
Je suis aux États-Unis -> J'y suis
Je suis en France -> J'y suis

En -> Of them (NOT TO BE CONFUSED WITH in a feminine country - En France), replaces numbers or DE
J'ai deux pommes, j'en ai deux
I have two apples, I have two of them
Je n'ai pas de pommes, je n'en ai pas
I don't have apples, I don't have them

Lui
INDIRECT (à) SOMEONE
À someone (he or she)
Je parle à Louis / Je parle à Marie
Je lui parle/I speak to him or her
There is NO difference between him/her in this context

Leur (à multiple people)
Je parle à mes parents, je leur parle
I speak to my parents, I speak to them

Le/La/Les
DIRECT OBJECT (no à)
Je mange le croissant/Je le mange
Je mange la baguette/Je la mange
Je manges les croissants/Je les mange
Je manges les baguettes/Je les mange

Le/La/Les in direct context can be to people if no à is involved
Je connais John Cena(M)/Je le connais
Je connais Céline Deon(F)/Je la connais
Je connais Les Beatles/Je les connais
