The answer is simple: Qui notes the subject of a relative clause and que notes the object of a relative clause.
Le professeur est mon voisin.
Here, "le professeur" is the subject of the sentence. If I want to make it the subject of the relative clause "est mon voisin", I use "qui", so we get :
Le professeur qui est mon voisin.
Now let's try a different sentence:
Je connais le professeur.
Here, "le professeur" is the object of the sentence. If I want to make "le professeur" the object of the relative clause, I use "que", so we get:
Le professeur que je connais.
