Make it descriptive, including relevant context, but also to the point. This way you improve your chances of getting a more relevant and specific answer.

You don't add -s in -er verbs (and some other verbs such as aller) in the second person singular UNLESS you have the pronoun y or en next to it.

Yep, some -ir verbs are special in that they conjugate like -er verbs in the present. Thus they undergo the same rule in the imperative.
Cueillir, offrir, souffrir, etc.

the thing that makes it one thing is "un/une" so the sentence you wrote is correct, if you're talking about one letter and if you've mentioned that letter already
« en … [chiffre] » is the structure
i'd say usually yes, because full stop is used for undefined plural (if that makes sense, not sure about my translation lmho) but it can also used for nouns that are uncountable e.g. sugar
il veut du sucre --> donne-lui en
or in your case
il veut une lettre --> envoie-lui en une
il veut des lettres --> envoie-lui en
