if your trenches/horizontal tunnels are at least 3 blocks wide/3 blocks high then you shouldn't need to place any blocks

just put rope on the crimson sides so thorns don't spread over

and planter boxes/platforms on the roofs of the tunnels so vines don't grow down

all are valuable, but you can minmax to create a good set up. I personally value defense the highest then go down > regen > dodge chance > damage reduction. Of course this all depends on the scenario too, if your enemy has very high damage you'd want to prio damage reduction first but usually you can meet your defense points to the enemies damage so you don't need damage reduction
