There is "the merchant", which is a specific NPC but there's several others that either sell things or have specific functions. You have to keep building houses for them and meeting they requirements for them to move in

I'll tell you a good template, I think it should work and keep you motivated.
First, just look up a list of bosses in order of progression.
Then just keep playing and exploring to the best of your abilities until you feel you're ready to give a shot to the first one. If you do not succeed, just try to think about what could be better. Maybe you were too slow, maybe the boss hit you for too much damage and you're lacking defense, or maybe you don't have a good weapon so you just ended up dying because you can defeat it fast enough

After that work your way the list, always thinking of what you can do to improve

And always read the wiki on things you don't know what it does or how to get it

Yes, absolutely. Focus your loadout on piercing, crowd control and long range weapons to deal with the Destroyer, Probes, Prime limbs and Twins efficiently. Make sure to kill Destroyer first and do not kill Prime until the other Mechs are down, as that will free them. Mechdusa also has less health than the 3 Mechs would have combined, and it can't move as freely as they would in Mecha Mayhem.

Great options include Dart Rifle/Pistol with Cursed/Crystal Darts, Shadowflame Knife, Spirit Flame, Orange Zapinator, Stormbow with Unholy Arrows, Onyx Blaster with Crystal Bullets, Bananarang and Drippler Crippler.
