Yes, but it’s not recommended to use a SD/Micro SD storage of more than 128GB on a 3DS as doing so will cause certain issues with your system.

Also, you would need to format any SD/Micro SD over 32GB to FAT32 before using it on a 3DS.

About SD-Card formatting

The 3DS family can only read SD-Cards if theyre formatted in FAT32.
For cards under 32GB this can be achieved with any standard tool.
For cards above 32GB on Windows you will need a special tool,
which can be downloaded here.
64GB cards need an Allocation unit size of 32KB/32768 bytes,
128GB need 64KB/65536 bytes.
Cards above 128GB are not recommended because of performance issues.

If your SD card is already FAT32, then you don’t have to format it.

As for moving files, I don’t think you have to. However, I advise making backups of all your files in case of any problems along the way.
