Ectoplasm shows up post-Plantera, and we know that Plantera is an amalgamation of many souls, not unlike Polterghast.

After Plantera was destroyed, the spirits within it were released and they move to the dungeon, which is why Dungeon Spirits show up and drop ectoplasm.

The spirits inside Polterghast are much angrier than the ones in Plantera (because Yharim), so that explains why the necroplasm is basically "infected" with malice unlike the more pure ectoplasm.

With this in mind, it is possible that necroplasm and ectoplasm can be created in-lore by getting a bunch of spirits (I'm thinking it's like squeezing juice out of a lemon). That's probably how the Onyx Kinsmen managed to get necroplasm to make Statis' Ninja Belt.
