When that castle was made "non hostile" after you make an ally and meet him there, you can't access the chest.

as long as you have free reign and you arent too far ahead in the story (meeting Mitsuhide) then you can get the chest

#

if not then its probably bugged and you will need to get past act 1 to get the chest
