Basically;
Story express = Gives you a couple goodies and already enhanced/honed/upgraded equipment so you can just focus on the story, and that goes all the way to the start of the endgame, BUT you have to start literally from 0 on a new character, so its all the story all over again.
Island speedrun = Will take longer and you actually have to hone the armors you get to clear the next continents, but the island route will give you most, if not all the materials you need for that, it takes a couple days to complete jumping from island to island and doing all sort of quests.

I wanna say that, in the long run, Story express on a new char could be faster overall due to you not needing to hone any equipment at all, due to the event giving you all you need to advance, if you dont mind doing it all again (Now that we have Shift+G, a way to fast skip dialogues from quests you already cleared, it will be a lot faster for you to clear up until shushire, will save you some hours of G mashing)@haughty ravine

Both of these options will provide you with enough materials to hone your gear all the way up to the start of the endgame right now (ilvl 1370), provided that you only open what you need from the chests that you can select stuff from
Don't blindly open the selection chests unless you know you want that specific thing, many beginners make this mistake (especially with battle item selection chests)
