if you suddenly lost 500 friends then theres some possibilities of what happened
- they all unfriended you en masse for whatever reason
- some sort of error occurred that caused the friend status to get corrupted or to not be displayed correctly (unsure of this)
- those 500 people opted to delete their account (also unsure of this, mainly because i dont know how this stuff works under the hood and the unlikelihood of people just up and deleting their accounts out of the blue)

if you cant add someone then its likely that they have blocked you and that prevents you from sending a friend request

software usually cache data to bother the api less often
give the game a restart and try adding someone
