I'm not sure i understand your question but, lighting up the campfire at the end of Alpine counts you in Caldera, lighting up the campfire at the end of tropics counts you in Alpine, etc etc

you're counted in the biome as soon as you light up the campfire right before it

basically, you're counted in the tropics when you light up the campfire at the end of the shore

Shore: ||Start by heading left and looting all the cases along the way. Begin your climb near the middle-right side of the mountain. You’ll notice three to four bridges grouped together in that area. Climb up toward the bridges, then continue upward and to the right until you reach the summit.||

Tropics: ||Make your way to the center to start climbing. Early on, you’ll come across two waterfalls. Head toward the left one. At this point, you’ll find two options. One path goes right toward the second waterfall, where you’ll find a long vine that takes you high up. This shortcut is great if you have the stamina for it. The other path keeps you climbing near the left waterfall and gradually leads you upward.

Both paths eventually bring you to a huge tree on the left side of the map. If you took the shortcut on the right, keep climbing up until you reach the summit. If you followed the left path, climb to the top of the large tree, then continue going up and to the right until you hit the summit. The right-side shortcut is quicker but uses more stamina, while the left side is a safer climb overall.||

Alpine: ||Head to the right side of the map. You’ll spot three luggage pieces and a shelf fungus just before reaching the base of the mountain. Start climbing on the right side. From below, you’ll be able to see a tree high up on the slope. If you have the stamina, this route is smooth and leads straight to the summit.

If not, stay along the right side and work your way up and slightly left. Eventually, the path might look like a dead end, but keep going. You’ll come across a lava spill and a few trees. If you see both, you’re in the right spot. From here, two paths open up. One to the right leads into a cave but also has a way around it. The other to the left is the safest option to reach the summit.||

Kiln: ||Today’s climb on Kiln was a bit tougher. I tried to go item-light to test out the best route. As usual, there’s a patch of firerock early on. This one is very safe and even has a nice ledge to rest on. If you’re bringing climbing gear, Kiln will be a breeze today.||

I don’t think I’m meant to do today run in the timed and ascent 1 ; the first time in tropics I was climbing a vine and it started raining which caused my stamina to waste once getting to the climb point. The second time I didn’t have enough stamina in tropics or the same items , so climbing that same vine resulted in my death. The third time I had the right stamina and items but when unequippimg my bag to get my energy drink on a standing point the bag disappeared :(

Mushrooms spawn in set locations, and it's rng for what mushroom it is and if they're poisonous or not, I believe. (I think it's also rng if they spawn at all lol)
