Honestly best way to know is sim without and then hit a target dummy till your DPS seems within standard deviation of sim for atleast 5 minutes.

To upgrade the trinket, you have to go to the Quartermaster, and do the weekly quest to get a currency item which allows you to interact with a forge to spend points on a talent tree. You also get a currency item from doing the singularity event. The first 4 points highlighted here provide buffs while inside the Voidspire.
