#Are the test subjects from OE screaming from getting blended?

it is entirely up to speculation!!
I highly doubt they really thought that far ahead to OE back in S1, nor does seemingly anyone on staff know besides whoever was in charge of making the soundscape in the first place.
From a Eurogamer interview with S1’s co-director, Tsubasa Takaguchi:
There's one mystery that's been baffling players since Splatoon's release: upon completing the game, and defeating its challenging final boss, those that stick around can hear distant, tortured screams. What on earth could it mean? Sakaguchi is just as puzzled as everyone else. “It's the first time I've heard this! I was the one who was in charge of the mechanics and the effects for the final boss. Towards the end, though, I was running out of time so I was asking the staff around me to do things for me - like the staff credits and many other things. Maybe it's a curse from those other staff. The final boss was one of the last things that we were working on. We were all high on adrenaline. Maybe there's some element of that in there, of that energy in there. Once I go back to Japan, I'll play it and have a look.”
Believe and speculate all you wish about this track, but to be honest it’s perfectly possible it’s merely some unsettling ambience a sound designer made for kicks

The bossfight is over “The Child Making Pit” where octarian couples go to slice off eachothers tentacles to create a child, and the screams are screams of pain, and adrenaline, as they fight over who must sacrifice a limb with comically large scissors
