Not sure I understand your question. Coupons and promotion codes can be used in checkout for any customer that is buying something the coupon applies to.

Can you explain a bit more about why you are unsure they will apply in your scenario?

So, I want to give discount for customer to his first purchase and apply this automatically. And my question is can I apply this while I create the customer. Or I should apply this when create checkout session?

In that case I think you would want to have the customer put the promotion code in at checkout

Otherwise I am not sure if it would apply to the first payment/show in checkout though you can set a customer up like that to test the behavior
