Do I have to complete the Field Guide before I do the final Quest? or can I work on it after that?

If you're aiming for the final Quest, I would say focus on that. Also though some Field Guide pages offer XP and necessary unlocks.

Oh sorry, yes it's actually great to do the graduation first and
then finish up any remaining field guide pages at your own time.

The dialogue changes for the final quest depending on how much of your Field Guide you've done, but it only detects Challenges; if you've finished every challenge but still have some collections to fill in, Professor Weasley will still say there's nothing else you need to do to complete your Field Guide
