You get the quest for Alohomora right after you complete the First Trial
You can fully unlock the spell right after you complete the main story quest, as long as you actually go to the effort to collect enough demiguise moons. There are enough in Hogwarts, Hogsmeade, and the nearby hamlets to get it to tier 3 without having to go to the second half of the map.
