Currently if your truck is in your base you cannot assign it to an expedition, you have to have it leave the base, then you can assign it, and it then has to drive back in (if it needs to load drones/cargo). This seems a little redundant and a waste of time. It would be nice to be able to assign missions to the rovers that are idle in base.

You should be able to do this with the expedition button on the bottom left of the global map. I have been assigning missions to trucks inside the base since I started playing the game. Select mission click on the truck section and pick a truck/rover.
