No, you wouldn't use Replace Color. And don't need the "greyscale" texture here.
Should just need to input the texture you want to swap colours into the UV port on the the gradient texture sample.
Also change Wrap mode to Clamp in the gradient texture import settings.
May also need sRGB disabled on the original texture.
