on a 10 def boss a 10% will make it so the boss divides your dmg by 9 instead of 10.

on a 25def boss (ex: nm150+ bosses) it changes to be like a 22.5 defense.

most extreme boost should be on co op slimes which have a 10k defense which in such a case goes down to 9k iirc.

imo hitting at least the 10% mark should be crucial to truly using this.
