Threadlings were never generated from damage, only through abilities and exotic armor/weapon perks.
I don't know what you're referencing with "it" in "Does it have to be strand?". Are you asking if your super has to be Strand?
And the question of using a Strand Weapon doesn't come into play because the aspect only talks about using your Class Ability to generate Threadlings
