I am so confused, does the last mote only drop from the Meatball in the blooming?

it can drop from any of the bosses

I have 16 motes, I have all the traveler balls placed, I can see the chest. I have the Legendary version of the gun. Do I just spam Overthrow and pray the mote drops?

yes, just kill each boss until it drops

Any idea if it makes a difference if the Motes are in my vault?
